Project description:
Transcription factor Nrf2 regulates a battery of genes encoding carcinogen-detoxifying enzymes. The Keap1/Nrf2 complex is vital in regulating free Nrf2 available for activation of detoxifying enzymes. Small molecules that selectively modulate the Keap1/Nrf2 interaction would be of great value in dissecting the functional aspect of Keap1/Nrf2 interaction in cells. The selectivity of small molecule is essential. The first specific aim of this project is to develop a novel structure-based molecular modeling methodology to design selective chemical modulators of Keap1/nNrf2 interaction. Next objective is to extend the methodology for the general application in selectively modulating specific protein-protein interactions by chemical approach.
